BMSCs promote the differentiation of NSCs into oligodendrocytes via mediating Id2 and Olig expression through BMP/Smad signaling pathway

Abstract: Neural stem cells (NSCs) have emerged as a promising treatment for spinal cord injuries. However, the increasing expression of bone morphogenetic proteins (BMPs) in spinal cord injury lesion sites seems to have contributed to the limited oligodendroglial differentiation and the majority of the astroglial differentiation of NSCs. In the present study, we demonstrate that BMPs promote NSCs differentiation toward astrocytes and prevent them from differentiating into oligodendrocytes. This effect is accompanied by… Show more

“…In the process of functional reconstruction aer SCI, neurons subsequently differentiate into functional neurons and play a role in signal transmission, whereas oligodendrocytes promote the regeneration of the myelin sheath and axons, which exert a positive effect on functional recovery. 45,46 The results of cell differentiation assays were consistent with the expected results for spinal cord recovery, suggesting that PLGA/FTY720 exerted a positive effect on functional reconstruction. Although qPCR and immunouorescence staining did not reveal obvious effects of PLGA/FTY720 on NSC differentiation into neurons, the proper mass ratio of PLGA/FTY720 nanobres induced NSC differentiation into oligodendrocytes, which promote axon regeneration.…”

“…Neural stem cells (NSCs) existing in the subventricle zone of the brain's germinal region and the subgranular zone of the hippocampus are capable of self-renewing, proliferating, migrating, and differentiating into various types of cells within the brain and spinal cord tissue [13,41]. NSC differentiation into neurons, astrocytes, and oligodendrocytes could replace necrotic cells resulting from injuries to promote the structural and functional repair of the brain and spinal cord [50].…”

“…MSCs were cultured following the procedure described in our previous studies [27,28]. The cells were isolated from the bone marrow of Fischer 344 rats and were cultured in DMEM (low glucose, Hyclone) containing 10% fetal bovine serum (FBS) (Gibco, USA) and 1% antibiotic solution at a density of 1 × 10 6 cells/cm 2 .…”
