Ca2+/calmodulin-dependent protein kinase kinase-β acts upstream of AMP-activated protein kinase in mammalian cells

Abstract: AMP-activated protein kinase (AMPK) is the downstream component of a kinase cascade that plays a pivotal role in energy homeostasis. Activation of AMPK requires phosphorylation of threonine 172 (T172) within the T loop region of the catalytic alpha subunit. Recently, LKB1 was shown to activate AMPK. Here we show that AMPK is also activated by Ca(2+)/calmodulin-dependent protein kinase kinase (CaMKK). Overexpression of CaMKKbeta in mammalian cells increases AMPK activity, whereas pharmacological inhibition of C… Show more

“…Recently neuronal over-expression of the Ca 2+ /calmodulin dependent protein kinase (CaMKK) was shown to protect mice from high fat diet-induced weight gain, insulin resistance and glucose homeostasis. CaMKK is activated by the change of the intracellular Ca 2+ [62-64] and acts as an upstream activator of AMPK, independent of AMP levels. In hypothalamic neurons, CaMKK regulates orexigenic NPY production [65, 66].…”

“…The catalytic α subunit contains a conventional serine/threonine protein kinase domain, and phosphorylation of the Thr 172 residue within the activation loop of the α subunit by upstream kinases is essential for the activity of the heterotrimer (reviewed by Carling [3]). Two kinases have been established as upstream activators of AMPK: the protein kinase serine/threonine 11 (LKB1)/ STE20-related kinase adaptor alpha (STRAD)/calcium binding protein 39 (MO25) complex [4] and the calcium/ calmodulin-dependent protein kinase kinase β (CAMKKβ) [5]. Once phosphorylated at Thr 172 , AMPK can further be activated by allosteric binding of AMP to the regulatory γ subunit [6].…”

“…Thr-172 can also be phosphorylated by the Ca 2 + -activated kinase calmodulindependent kinase kinase-b, which can trigger the activation of AMPK in response to agents that increase intracellular Ca 2 + (Fig. 1), even in the absence of a rise in AMP (8,9) . AMPK occurs universally as heterotrimeric complexes, i.e.…”
