Calcium Binding Rigidifies the C2 Domain and the Intradomain Interaction of GIVA Phospholipase A2 as Revealed by Hydrogen/Deuterium Exchange Mass Spectrometry

Abstract: The GIVA phospholipase A 2 (PLA 2 ) contains two domains: a calcium-binding domain (C2) and a catalytic domain. These domains are linked via a flexible tether. GIVA PLA 2 activity is Ca 2؉ -dependent in that calcium binding promotes protein docking to the phospholipid membrane. In addition, the catalytic domain has a lid that covers the active site, presumably regulating GIVA PLA 2 activity. We now present studies that explore the dynamics and conformational changes of this enzyme in solution utilizing peptide… Show more

“…The change in exchange upon binding to 500 M lipid vesicles was tested in the presence of 0, 200, and 1000 M Ca 2ϩ . Significant decreases upon exposure to the lipid sur- (35). A significant decrease in exchange due to binding of lipid vesicles was seen in the region 36 -39, as well as a small change in the region 28 -35 in the presence of 1 mM Ca 2ϩ and PAPC lipid vesicles (Fig.…”

“…This approach has traditionally been used to study protein-protein and protein-ligand interactions, but we applied it to look at the interaction of water-soluble proteins with membranes and demonstrated this approach with PLA 2 (136 -138). We have now studied four of the major types of PLA 2 , including the secreted (139), cytosolic (140,141), and Ca 2ϩ -independent forms (142), as well as lipoprotein-associated/platelet-activating factor (PAF) acetyl hydrolase (143) and the interactions of the latter with lipoproteins such as HDL (144). These DXMS methods also led to a better understanding of the catalytic action of an unusual form with a Cys in the catalytic site (145).…”
