Carbohydrate metabolism is a determinant for the host specificity of baculovirus infections

Abstract: Summary Baculoviruses Autographa californica multicapsid nucleopolyhedrovirus (AcMNPV) and Bombyx mori nucleopolyhedrovirus (BmNPV) have highly similar genome sequences but exhibit no overlap in their host range. After baculovirus infects nonpermissive larvae (e.g., AcMNPV infecting B. mori or BmNPV infecting Spodoptera litura ), we found that stored carbohydrates, including hemolymph trehalose and fat body glycogen, are rapidly transformed into glucose;… Show more

“…At the same time, by annotating the KEGG pathway of DEGs, we found that DEGs were mainly related to amino acid metabolism and carbohydrate metabolism in the metabolic category (Figure 2b). Studies have shown that the amino acid content in the haemolymph of silkworms infected with BmNPV will change significantly (Feng et al, 2021), and the content of carbohydrates (such as trehalose and glycogen) will also change (Tsai et al, 2022), which is consistent with our results.…”

“…The approach may also apply to other baculoviruses which have a wide host range. The baculovirus AcMNPV is known to infect larvae of more than 39 species of Lepidoptera insects, including Plutella xylostella, Trichoplusia ni, and Spodoptera frugiperda [51,52]. Many variants of AcMNPV have been isolated from Lepidoptera insects besides A. californica, such as Plutella xylostella multiple nucleopolyhedrovirus isolate CL3 (PlxyMNPV-CL3) isolated from P. xylostella [53], RoMNPV isolated from Rachiplusia ou [54], and AfMNPV isolated from A. falcifera.…”

“…As the center of intermediates and energy metabolism, the TCA cycle occupies a critical position in various metabolic pathways [ 38 ]. A previous study showed that AcMNPV infection upregulated the expression of metabolic enzyme genes ( cs , aco2, and idh3 ) in the host TCA cycle [ 10 ]. Therefore, we examined the transcription levels of these three genes in BmN cells after BmNPV infection.…”

“…It is also the final metabolic pathway for glucose, protein, and lipid [ 9 ]. The study of energy metabolism in Autographa californica multiple nucleopolyhedroviruses (AcMNPV) showed that viral infection enhances the consumption of glucose in the host, which then enters the glycolytic pathway and TCA cycle [ 10 ]. In addition, AcMNPV infection drives a large amount of ATP synthesis by activating the expression of TCA cycle genes [ 11 ].…”
