Cell injury releases endogenous adjuvants that stimulate cytotoxic T cell responses

Abstract: General immunostimulants (adjuvants) are essential for generating immunity to many antigens. In bacterial infections, adjuvants are provided by components of the microorganism, e.g., lipopolysaccharide. However, it is unclear what provides the adjuvant effect for immune responses that are generated to tumors and many viruses. Here we show that cell injury and death of tumor or even normal cells provide a potent adjuvant effect for the stimulation of cytotoxic T lymphocyte responses. This adjuvant activity is c… Show more

“…In support of this notion, cytosol obtained from healthy cells has adjuvant activity, indicating that DAMPs are present intracellularly 11 . It isn't yet clear whether there are also membraneassociated DAMPs, but nuclei have little adjuvant activity 11 . In living cells, these intracellular molecules are normally sequestered from the innate immune system by the plasma membrane.…”

“…When healthy cells were treated with protein synthesis inhibitors, their endogenous adjuvant activity was not reduced 11 . Moreover, when healthy cells were killed instantaneously by freeze-thawing, they provided adjuvant activity 10 11.…”

“…Given the large body of evidence supporting the adjuvant effect of tissue damage (11,15,(17)(18)(19)(20)(21), it is surprising that CTL-mediated tissue destruction has such a minimal immunological impact. Moreover, the CTLs themselves should produce inflammatory cytokines, which might be expected to promote immunity.…”

“…While CTLmediated tissue destruction will release self-antigens through target cell apoptosis, this cell death may be accompanied by the production of CTL derived inflammatory cytokines such as TNF␣ and IFN-␥ (16) or with the release of inflammatory components of the degraded extracellular matrix such as heparan sulfate and hyaluronan oligosaccharides (17,18). Depending on the extent of tissue destruction, secondary necrosis may also accompany CTL damage, leading to the release of intracellular components of potential immunogenicity (19)(20)(21). Collectively, these signals may be sufficient to overcome the immunosuppressive nature of apoptotic material and drive T cell priming against any released antigens.…”

“…This leads to the spillage of cellular contents in the extra-cellular milieu which might provoke inflammatory reactions in neighbouring cells (Matzinger, 1994;Shi et al, 2000). Here, presence of GbpA was found to increase cell membrane permeability in host cells which is indicative of cell necrosis.…”
