Cell-penetrating peptides as a promising tool for delivery of various molecules into the cells

Abstract: Many biologically active compounds, including macromolecules that are used as various kinds of drugs, must be delivered to the interior of cell or organelles such as mitochondria or nuclei to achieve a therapeutic effect. However, very often, lipophilic cell membrane is impermeable for these molecules. A new method in the transport of macromolecules through the cell membrane is the one based on utilizing cell-penetrating peptides (CPPs). Invented 25 years ago, CPPs are currently the subject of intensive resear… Show more

“…TP reveals some characteristic features of both galanin and mastoparan since it inhibits the binding of galanin to GALR-1 receptor as well as modulates the activity of Gs proteins due www.fhc.viamedica.pl to the inhibition of GTPase activity [9]. The shorter analogs of TP (TP7-15) showed lower cytotoxity in comparison to TP, galanin and mastoparan [8].…”

“…[57] Up to 30 amino acid residues can comprise one CPP, of which many are arginine or lysine. [58] Although research is still being conducted concerning how CPPs enter cells to deliver their cargo, two major cellular uptake mechanisms are known: energy independent pathways and endocytosis. Examples of models that have been suggested for the latter include the inverted micelle, pore formation, carpet, and membrane thinning models.…”

“…Recombinant proteins are usually considered as extracellular antigens; however cell penetrating peptides and their cargos can traverse the cytoplasm membrane [10,11] and can be considered as intra cellular antigens. Recently, we reported an expression system for are combinant PTD-J-cargo protein comprising a protein transduction domain (PTD), a J domain of Hsp40, and a cargo polypeptide [12].…”
