Challenges of Current Anticancer Treatment Approaches with Focus on Liposomal Drug Delivery Systems

Abstract: According to a 2020 World Health Organization report (Globocan 2020), cancer was a leading cause of death worldwide, accounting for nearly 10 million deaths in 2020. The aim of anticancer therapy is to specifically inhibit the growth of cancer cells while sparing normal dividing cells. Conventional chemotherapy, radiotherapy and surgical treatments have often been plagued by the frequency and severity of side effects as well as severe patient discomfort. Cancer targeting by drug delivery systems, owing to thei… Show more

“…According to the ACS, an estimated 608570 Americans were anticipated to succumb to cancer in 2021, equating to ≈1670 deaths daily and accounting for a quarter of total deaths in the United States of America. [ 15 ] These figures underscore the urgency of anticancer research and the imperative need for pioneering treatment approaches.…”

“…Long-term exposure to cisplatin can result in renal dysfunction, characterized by nuclear pallor and mitochondrial swelling, while Anthracyclines like doxorubicin can cause acute cardiotoxicity, such as arrhythmias and myocarditis, due to the accumulation of free radicals. [15] Improving the effectiveness of cancer chemotherapy stands as a significant challenge for scientists in the field of clinical oncology. Studies reveal that a modest percentage, specifically 30-40%, of individuals undergoing antitumor chemotherapy witness significant advantages.…”

“…These challenges have attracted the application of nanotechnology to improve its success and sidestep impacts on patient survival. [3][4][5][6] The upcoming application of nanotechnology in medicine and healthcare (nanomedicine) has gained popularity. Predominantly, its potential in drug delivery and medical diagnosis has notable hope to prevent and treat severe conditions such as cancer.…”

“…Polymers are able to reinforce liposome structures by providing steric hindrance and are thereby able to prevent liposome fusion and inhibit protein opsonization (Yan et al 2005;Gyanani et al 2021). Hydrophilic polymer chains can be covalently conjugated to either the head group or chain of the lipids followed by liposome formation (Huang et al 2020).…”
