Chemical Composition of Miscanthus sinensis var. purpurascens Flower Absolute and Its Beneficial Effects on Skin Wound Healing and Melanogenesis‐Related Cell Activities

Abstract: Miscanthus sinensis var. purpurascens (MSP, flame grass) is found in Korea, Japan, and China, and its biological activities include anti‐cancer, detoxifying, vasodilatory, antipyretic, and diuretic effects. However, no study has investigated the effects of MSP on skin‐related biological activities. In this study, we explored the effects of the absolute extracted from the MSP flowers (MSPFAb) on skin wound healing‐ and whitening‐related responses in keratinocytes or melanocytes. MSPFAb contained 6 components an… Show more

“…Woodley et al reported that type I and type IV collagens increase keratinocyte migration [ 49 ]. Previous studies have shown many plant extracts enhance the syntheses and secretions of type I and/or IV collagens by HaCaT cells [ 25 , 30 , 50 ], which implies keratinocytes can synthesize and secrete collagen types I and IV. In the present study, SKAFAb increased collagen type I at concentrations of 150 μg/mL and 50 and IV syntheses 150 μg/mL in HaCaT cells.…”

“…B16BL6 cell proliferation was assayed using a BrdU proliferation ELISA kit (Roche, Indianapolis, IN, USA) [24,51]. Briefly, 2 × 10 3 cells were seeded in each well of 96-well plates and cultured for 12 h. Different concentrations of SGMFAb in MEM containing 0.2% DMSO with or without 2% FBS were added to each well, followed by incubation for 36 h. The cells were labeled in BrdU-labeling solution (10 µM) for 12 h at 37 • C, and the culture medium was then removed.…”

“…Tyrosinase activity was analyzed based on the dopachrome method using L-DOPA as substrate, as reported previously [24,51]. In brief, B16BL6 cells (1 × 10 5 cells/dish) were cultured in 60 mm culture dishes for 12 h and then incubated in MEM (with 2% FBS) in the presence or absence of SGMFAb at different concentrations with or without α-MSH (200 nM) for 48 h at 37 • C. Cells were then lysed and centrifuged as in the melanin content assay described above.…”
