Chitosan‐Based Nanocoatings for Hypothermic Storage of Living Cells

Abstract: The formation of ultrathin chitosan-based nanocoating on HL-60 model cells and their protective function in hypothermic storage are presented. HL-60 cells are encapsulated in ultrathin shells by adsorbing cationic and anionic chitosan derivatives in a stepwise, layer-by-layer, procedure carried out in an aqueous medium under mild conditions. The chitosan-based films are also deposited on model lipid bilayer and the interactions are studied using ellipsometry and atomic force microscopy. The cells covered with … Show more

“…For example, cationic polymers have been applied as gene carriers in nonviral gene therapy [ 48 ] and as drug delivery systems [ 49 ]; their potential use as anticancer drugs has also been suggested [ 50 ]. Moreover, a hydrogel containing GTMAC-modified polysaccharide can be applied to cell encapsulation and protein delivery [ 51 , 52 ]. Based on our results we can claim that high molecular weight linear polymers perform better as therapeutics than the cyclic or low molecular weight cationic polysaccharides.…”

“…14 In brief, aliquots of 10 islets (free or encapsulated) per well in triplicate 96-well plates were incubated in culture medium. We added 5 g/L MTT solution to culture medium at 1:10 dilution, which was followed by 2-hour incubation.…”

“…The commercial and laboratory chitosan products are not completely deacetylated and tend to be copolymers of N-acetyl glucosamine (NAG) and N-glucosamine [3][4][5] . The ratio of these two repeating units depends upon the source and method of preparation of chitosan but glucosamine unit predominates 6,7 .…”
