Chloroplast Division and Expansion Is Radically Altered by Nuclear Mutations in Arabidopsis thaliana

Abstract: We have isolated three mutants of Arabidopsis thaliana in which there is a sevenfold change in chloroplast number in fully expanded leaf mesophyll cells and increases and decreases in chloroplast number are compensated for by changes in chloroplast size. The changes are stably inherited in sexual crosses for three generations and mutant phenotypes are effected by changes at single recessive nuclear loci, termed arc loci. This is the first report of large, stably inherited changes in chloroplast number in highe… Show more

“…Although a dozen of Arabidopsis arc mutants relevant to the chloroplast division process have been reported many years ago [35,36], only a few of them are cloned by map-based cloning during a long period of time [22,23,[37][38][39]. Mapping genes for these mutants is laborious, because one has to use microscope for phenotyping [22,38].…”

“…The small but statistically significant decrease (Student's t test, P > 0.01; less than one chloroplast per optical section) in chloroplast number seen in the msl2 msl3 arc11 and msl2 msl3 arc3 triple mutants is likely due to the variability in cell size seen in the msl2 msl3 mutant background. Cell size is linearly correlated with chloroplast number (Pyke and Leech, 1992).…”
