Keratinization defects can be very severe and disfiguring diseases. The development of retinoids such as etretinate has provided us with an effective symptomatic form of oral therapy for these skin conditions. Based on our own experience, we briefly outline the therapeutic potential of etretinate in various keratinization defects (lamellar ichthyosis, Netherton syndrome, Sjögren-Larsson syndrome, mal de Meleda and juvenile pityriasis rubra pilaris). The toxicology of etretinate is reviewed with special regard to the treatment of children. Bone changes such as premature closure of the growth line or other unacceptable side-effects have so far not been observed. Guidelines for patient selection and for the safe treatment of children are given.

Summary: Possible side effects of the retinoids etretinate and isotretinoin on the germinal epithelium were examined. 15 men recevietretinate and 13 isotretinoin. After 16 weeks of treatment at therapeutic doses, no changes were observed in fundamental spermatological parameters. On the basis of examinations and data in the literature, we hold the opinion that retinoid treatment is to be considered safe from the andrological viewpoint. Zusammenfassung: Die mögliche Nebenwirkung von Retinoid (Etretinat und Isotretinoin) auf das Samenepithel wurde an 15 mit Etretinat und an 13 mit Isotretinoin behandelten Männern untersucht. Im Verlauf der 16 Wochen dauernden therapeutischen Behand‐lung in entsprechender Dosis konnten in den grundlegenden Parametern keine Verände‐rungen beobachtet werden. Aufgrund der Untersuchungen und der literarischen Angaben sind wir der Meinung, daß die Behandlung mit Retinoid in andrologischer Hinsicht als si‐cher bezeichnet werden darf.
