Cold-Induced Changes in Plasma Norepinephrine, Epinephrine and Dopamine Concentrations in Patients with Raynaud's Phenomenon

Abstract: Summary:To investigate the role of the sympathetic System in Raynaud's phenomenon, which has not yet been elucidated, we measured the levels of norepinephrine, epinephrine and dopamine before, immediately and 30 min after the cold pressor test in plasma from 17 patients with primary, 6 with secondary RaynautTs phenomenon and 19 volunteers, matched for age and sex. Patients had significantly low baseline epinephrine (0.13 ± 0.02 vs 0.37 ± 0.04, nmol/1, p < 0.001, mean ± S. E.), but normal norepinephrine and dop… Show more

“…In our study, among the 15 patients with Raynaud's syndrome, only one was Val/Val, the other 14 was carrying at least one Met allele. Changes in circulating dopamine, which might be affected by COMT Val158Met polymorphism, were observed in a study about Raynaud's disease (Marasini et al, 1991). These results might indicate that dopamine plays a role in Raynaud's phenomenon.…”

“…This theory is consistent with RP being the result of an intrinsic problem within the blood vessels and not a result of a systemic or humoral problem. 36 For example, levels of neither epinephrine nor norepinephrine are elevated in RP sufferers, 37 , 38 and it is possible to induce an RP episode in a digit that has its sympathetic nerve output blocked; 26 , 39 facts that both allude to vascular disorder.…”

“…Future studies with atomoxetine should thus be performed in a larger number of subjects. Preliminary results in humans indicate that atomoxetine, as well as sympathetic activation by ketamine and the cold pressor test ( Marasini et al, 1991 ) reduce [ 11 C]ORM-13070 binding by 10–15% in human subjects ( Lehto et al, 2014 ; Scheinin et al, 2013 ).…”
