Combined Biochemical and Serological Typing of Clinical Isolates of Klebsiella

Abstract: In a series of 640 strains of Klebsiella isolated from clinical specimens over a 7-month period, there were sufficient biochemical differences between strains to allow a biochemical typing system to be established. Biochemical tests were done in solid media inoculated with a modified Steers inocula replicator. Biotypes were designated by a numerical coding system; 29 distinct biotypes were found among the 640 strains of Klebsiella. Serotyping of 270 of the strains was done by the Quellung reaction, and 40 caps… Show more

“…However, only a total of 39 serotypes occurred in common among K. planticola, K. oxytoca, and K. pneumoniae. Although there were reports that serotypes 2 and 21 strains occurred at frequencies significantly higher than did all other serotypes in the bacteremic isolates (4, 5), the majority of prior reports showed that no one serotype or serotypes were found to predominate in Klebsiella strains from various clinical sources (15,24,26). The results of the present study were in accord with this conclusion.…”

“…The end point was the reciprocal of the greatest dilution at which a positive reaction could still be clearly detected. Cross-reactions between several capsular types were eliminated by the titration, because the titer of a cross-reaction is lower than the homologous reaction (24,25).…”

“…Biotyping based on an extended panel of biochemical and culture tests is certainly the most practicable method of typing for smaller laboratories that are epidemiologically not optimally equipped. Biotyping can be carried out by using macrotube tests alone (100,202) or by combining a commercially available miniaturized system such as the API 20E system with additional macrotube tests (185,217). However, because of the large number of reactions to be tested and the often long cultivation times-up to 90 days for demonstration of gelatinase (228)-biotyping of Klebsiella spp.…”
