Comparative Proteomics of Whey and Milk Fat Globule Membrane Proteins of Guanzhong Goat and Holstein Cow Mature Milk

Abstract: Guanzhong goat and Holstein cow milks are the major milk supply for the Chinese dairy industry. Whey proteins and milk fat globule membrane (MFGM) proteins of both milk were characterized and compared using proteomic techniques. A total of 283, 159, 593, and 349 proteins were identified, respectively, in whey and MFGM for the two species using Liquid Chromatography combined with Tandem Mass Spectrometry (LC-MS/MS). Functional categories analyses showed that both goat and cow MFGM proteins had three most abunda… Show more

“…Final digested samples were filtered using a 10 kDa ultra‐centrifugal filter to obtain the peptide mixtures, followed by desalting using a C18 stage tip. The desalted peptides were separated by Thermo Fisher easy nano‐LC 1000 system (Thermo Scientific) and characterized on a Q‐Exactive (Thermo Scientific) as previously described . In brief, the desalted peptides were loaded on a pre‐column (100 μm × 2 cm, 5 μm, C18 particle) and followed by an analytical column (75 μm × 100 nm, 3 μm, C18 particle).…”

“…This important information helped identify the main proteins with immunity-promoting properties for newborns [66]. These analyses have been performed to study and compare the proteome of various MFGMs, mainly from humans and cows [64][65][66], but also from other species such as the goat, yak, buffalo, horse, and donkey [61,62,[67][68][69][70].…”

“…In a 2019 study, a total of 776 MFGM proteins were identified: 427 and 183 that are unique for goat and cow milk, respectively, and 166 proteins shared between the two species. Most of the goat MFGM proteins were related to metabolic processes (about 21%), whereas most of the cow MFGM proteins were related to disease-associated pathways (about 49%) [62]. Subsequently, the same authors evaluated the variations between goat colostrum and mature MFGM proteome.…”
