Cost-Effective Recovery and Purification of Polyhydroxyalkanoates by Selective Dissolution of Cell Mass

Abstract: Highly efficient separation and purification of polyhydroxyalkanoates (PHAs) from PHA-containing cell mass is essential to production of the bioplastics from renewable resources in a cost-effective, environmentally friendly way. Based on selective dissolution of non-PHA cell mass (NPCM) by protons in aqueous solution and crystallization kinetics of PHA biopolymers, a simple process is developed and demonstrated to recover PHAs from cell mass to high purity (>97 wt %) with high yield (>95 wt %). The average mol… Show more

“…The lowest amounts of biomass showed by soil isolate S3 that was 87 mg and 41 mg in NA and nutrient broth, respectively. [15,16] Soil isolate S4 showed 232 mg in NA, respectively. By considered other view of production standard PHB producer A. latus gave yields 370 mg of biomass which was greater than biomass yielded by other soil isolates organism hence that standard organism was selected for further production.…”

“…The plates and broth were incubated at 30°C for 48 h. [15] Harvesting of cell mass Cell mass from agar plates was scrapped with the help of spatula and that from broth 5 ml of 48 h at 30°C individual grown isolate was centrifuged at 6000 rpm for broth culture and 4000 rpm for solid state agar medium at 10°C; centrifugation time 15 min was employed for both medium, these processes were carried out in cooling centrifuge (Remi, Mumbai). Then, obtained cell pellets were suspended in deionized water.…”

“…PHA determination was conducted according to the Comeou et al (1988) and PHA concentration was determined according to the method of Braunegg et al (1998) using benzoic acid as internal standard. CDW was determined as described previously by Wong (2001), and synthesis of PHA was analysis according to the method of Yu & Chen (2006).…”

“…Reduction of the molecular weight to some extent does not affect the mechanical properties of the biopolymers. The bioplastics, however, do become brittle or less ductile when the molecular weight is greatly reduced (e.g., <100-200 kDa) (Yu & Chen, 2006). PHA recovered by NPCM or dispersion digestion can change the molecular weight (M w ) of PHA.…”
