Curcumin Benefits as Antioxidant, Antiinflamation and Antiapoptosis Ameliorate Paracetamol Toxicity

Abstract: Background: Paracetamol poisoning due to the use of paracetamol overdose is the most prevalent case of poisoning. Toxic metabolites from paracetamol cause glutathione depletion and lead to hepatic cell death. Curcumin, a polyphenol substrat in Curcuma longa, has been known to ameliorate the toxic effects of paracetamol. The mechanisms have been known are curcumin as antioxidant, anti-inflammatory, and anti-apoptotic. The curcumin protection mechanism against paracetamol poisoning will be discussed. Methods: The… Show more

“…As the toxicity progresses, jaundice, coagulopathy, encephalopathy, and multi-organ failure may develop (7) . Chronic ingestion of supra-therapeutic doses can cause a subacute form of liver injury that may present with nonspecific symptoms, such as fatigue and malaise, and can progress to cirrhosis, end-stage liver disease, and kidney failure as well (12) . Laboratory findings that support the diagnosis of paracetamol toxicity include elevated transaminases (AST and ALT), bilirubin, prothrombin time, and declining bicarbonate (7) .…”

“…(Family: Zingiberaceae) has potential in the prevention and therapeutic interventions of several pathological conditions including respiratory diseases, inflammation, liver disorders and diabetic wounds. It prevents a variety of carcinogen-induced cancers in rodents by suppressing the mutagenic effects of carcinogens [1]. In addition, the hepato-and nephroprotective, thrombosis suppressing, myocardial infarction protective, hypoglycemic, and antirheumatic effects of curcumin are also reported [2][3][4].…”

“…Failure to comply with directions may lead to serious side effect (i.e., the correct dose, correct schedule, and for the intended disease). For example, overdose of acetaminophen has been associated with liver damage [1]. Analgesics increase the gastrointestinal tract bleeding even when taken at appropriate doses [2].…”
