Definition of new alleles of MIC‐A using sequencing‐based typing

Abstract: We have sequenced exons 2, 3 and 4 of MIC‐A in 23 homozygous cell lines, 22 families and 54 unrelated individuals. This has led to the definition of seven polymorphic positions in exon 2, 13 in exon 3 and 12 in exon 4, yielding a total of 33 different MIC‐A allelic specificities, of which 16 have not been described before. The newly defined sequences and those of the alleles defined before were entered into a database of the SCORE program (Helmberg et al., 1998, Tissue Antigens, 51, 587) for comprehensive geno… Show more

“…The reaction was performed at the annealing temperature of 65°C. Exons 2, 3, and 4 were sequenced separately using primers described previously (55). For ambiguous allele combinations, 2201-bp MICA PCR product from four donors were cloned using TOPO TA cloning kit for sequencing (Invitrogen Life Technologies) according to the manufacturer's recommendations.…”

“…Some of these associations are likely to be secondary associations with alleles of the closely linked HLA‐B locus. For the MICA gene there is evidence for very considerable polymorphism in exons 2–5 with a triplet repeat in exon 5 (14–20). The MICB gene is noticeably less polymorphic than MICA and contains a dinucleotide repeat polymorphism in intron 1 (20–25).…”

“…This new primer (5 0 -CAC CTG TGA TTT CCT CTT CCC CAG AGC) corresponds to sequence lying between nucleotide positions 6926 and 6952 in the reference MICA sequence (GI:1405892). The exon 2 forward primers used by others 2,17,18 are also outside the polymorphic region in intron 1.…”

“…Methods for genotyping include PCR-SSP, SSOP, RSCA, [10][11][12][13][14][15] along with sequencing-based typing (SBT) usually considered definitive. 2,[16][17][18][19] Results have differed for the same reference cell lines typed by alternative methods. As examples, the cell line WJR076 was typed as *017 by SBT 2 and PCR-SSP, 12 but also reported as *031.…”
