Determination of eight penicillins in serum from cattle and pigs by generic HPLC method

Abstract: KeyWordsColumn liquid chromatography Solid-phase-extraction Penicillin varieties Calf and pig serum SummaryAn HPLC method was developed for determination of amoxicillin, penicillin G, penicillin V, ampicillin, oxacillin, cloxacillin, nafcillin and dicloxacilhn in serum from pigs and cattle. Serum was cleaned up by solid-phase extraction (SPE), ultra-filtered and derivatised. The method was linear in the range tested up to 2000 ng mL-1 of individual penicillins in serum. Limits of detection were 11 -14 ng mL -1… Show more

“…Several analytical methods have been reported in the literature for the determination of Nafcillin and/or its related substances [ 11 13 ]. The USP has also published the monographs for both active pharmaceutical ingredients and formulation products and has given the methodologies for determining the assay of Nafcillin [ 14 ].…”

“…In the majority of papers, HPLC methods utilize UV detection at low wavelengths (f 230 nm) [9,10], although fluorimetric [8] and mass spectrometric detection [7] have also been reported. Sample preparation methods involve deproteinization and extraction techniques, such as LLE or for the most part SPE [1,5,6,13].…”

“…Further cleanup is achieved by column switching [6] and extraction techniques such as liquid -liquid extraction (LLE) [11,13] and SPE [4,16,18]. Restricted access materials (RAM) have been used by excluding macromolecules from the matrix while selectively retaining the hydrophobic micromolecules [12,20].…”
