Development and characterization of 32 microsatellite loci in Genipa americana (Rubiaceae)

Abstract: • Premise of the study: Microsatellite primers were developed for the tree species Genipa americana (Rubiaceae) for further population genetic studies.• Methods and Results: We identified 144 clones containing 65 repeat motifs from a genomic library enriched for (CT)8 and (GT)8 motifs. Primer pairs were developed for 32 microsatellite loci and validated in 40 individuals of two natural G. americana populations. Seventeen loci were polymorphic, revealing from three to seven alleles per locus. The observed and e… Show more

“…The total genomic DNA was extracted from approximately 100 mg of leaf material, according to the CTAB (cetyltrimethyl ammonium bromide) method described by Doyle and Doyle (1987), with modifications, namely: increased concentration of polyvinylpyrrolidone (PVP) from 1 to 2%; CTAB, from 2 to 5%; and βmercaptoethanol, from 0.2 to 2% in the extraction buffer; and incubation time reduction from 60 to 30 minutes (at 65°C).The DNA quality and quantification was evaluated by electrophoresis in a1% agarose gel stained with ethidium bromide (0.2 mg / mL) and by nanodrop. The quantified DNA was diluted to generate working solutions at 7.5 ng/μL (Manoel et al, 2014).…”

“…Microsatellite loci were amplified by polymerase chain reaction in a 15-mL final volume, using GoTaq ® Colorless Master Mix containing 7.5 mL 2X GoTaq ® Colorless Master Mix, 0.9 mM of each primer (forward and reverse), 3.0 μL nuclease-free water, and 7.5 ng template DNA. The amplification program for all the primers consisted of an initial denaturing step at 94°C for 1 min, followed by 35 cycles of each amplification at 94°C for 1 min, 1 min at the specific annealing temperature for each primer pair (Manoel et al, 2014), 72°C for 1 min, and a final elongation step at 72°C for 10 min. Amplifications were performed using a Mastercycler (Eppendorf, Hamburg, Germany).…”

“…Progeny and regenerant DNA was extracted from the leaf tissue of germinated seeds using the method of Doyle and Doyle (1987). Six primers were used (Manoel et al, 2014). Microsatellite loci were amplified by polymerase chain reaction in a 15-mL final volume, using GoTaq ® Colorless Master Mix containing 7.5 mL 2X GoTaq ® Colorless Master Mix, 0.9 mM of each primer (forward and reverse), 3.0 μL nuclease-free water, and 7.5 ng template DNA.…”
