Development of Human Monoclonal Antibodies to Rabies

Abstract: A total of nine human monoclonal antibodies (MAbs) to rabies virus were generated from peripheral B lymphocytes of subjects immunized with human diploid cell rabies vaccine by somatic cell hybridization. The MAbs were analyzed for their antigen-binding specificities using ELISA, Western blot, and immunoprecipitation assays. The different assays made it possible to identify MAbs directed to the surface glycoprotein, nucleoprotein, nominal phosphoprotein, and matrix protein. One of the MAbs that recognized the s… Show more

“…However, none of these approaches is likely to graduate to use in clinical settings in the near future. and to ensure in vivo protection in a Syrian hamster rabies challenge model; they have also been demonstrated to be effective in phase I trials in humans [129][130][131][132][133]. The successful development of such antibodies would help to ensure the supply of life-saving biological drugs to people exposed to rabies, at a reasonable cost.…”

“…L'utilisation des AcM murins est moins avancée et pose potentiellement des problèmes anaphylactiques (Muhamuda et al, 2007). La combinaison d'AcM humains présentant les mêmes capacités neutralisantes que les HRIG semble une bonne alternative (Dorfman et al, 1994 ;Champion et al, 2000 ;Bakker et al, 2005Bakker et al, , 2008Sloan et al, 2007). Ces anticorps sont sélectionnés pour présenter des spécificités et des fonctionnalités complémentaires.…”

“…Briefly, human rabies immunoglobulins (HRIG) are used in combination with rabies vaccination for a known or suspected rabies exposure, administered intravenously as well as directly into the suspected exposure site (see the chapter by T. Nagarajan et al, this volume). Native human antibodies have been cloned that are capable of neutralizing the virus in vitro and in vivo; most of these are reactive with the rabies glycoprotein (Dietzschold et al 1990;Gebauer and Lindl 1990;Lafon et al 1990;Ueki et al 1990;Enssle et al 1991;Dorfman et al 1994;T.
