Differential effects of glucose and fructose on hexose metabolism in dog spermatozoa

Abstract: ResearchIncubation of dog spermatozoa with 10 mmol l -1 glucose or fructose rapidly increased the intracellular content of glucose 6-phosphate and fructose 6-phosphate, although the effect of fructose was greater. These effects were correlated with increases in ATP, ribose 5-phosphate and glycogen contents, and in the rates of formation of Llactate and CO 2 . In all cases, except for ATP and glycogen, the effect of fructose was greater than that of glucose. The total hexokinase activity of the crude extracts o… Show more

“…Facilitated glucose transporters (GLUT) are among the candidates [34,35]. Various glucose transporter isoforms including GLUT1, 2, 3, 5 and 8 have been identified in testicular germ cells and spermatozoa in the rat, bull, dog and man [36][37][38][39][40][41][42]. In the report that the GLUT inhibitor phloretin inhibits osmotic water fluxes in ovine and human spermatozoa, it was suggested that some of these transporters may be involved in water transport [43].…”

“…The first point is the fact that the optimal utilization of sugars by sperm is reached to determined concentrations of this sugars. For instance, the optimal utilization rate of glucose by sperm from dog and boar is reached to very low concentrations of the sugar, at about 0.1 mM [16,36,37,45]. This indicates that the addition of low concentrations of sugar to the extenders would be enough to maintain sperm energy levels.…”

“…In fact, both fructose and sorbitol are typical vegetal sugars, without any significant presence in animals, excepting in mammalian seminal plasma. It is noteworthy that the utilization of sugars like fructose, sorbitol and mannose by sperm of species like boar and dog of is in fact less effective in order to obtain energy than that of glucose [36,45]. The greater effectiveness of glucose is mainly linked to a greater sensitivity to the hexokinase system, which phosphorylates sugars as a first step in their metabolization pathway [36,45].…”
