Differential expression of a poplar SK2-type dehydrin gene in response to various stresses

Abstract: Dehydrins are group II, late embryogenesis abundant proteins that act putatively as chaperones in stressed plants. To elucidate the function of dehydrins in poplar, we isolated the SK2-type dehydrin gene Podhn from Populus alba × P. tremula var. glandulosa suspension cells and analyzed its expression following treatments of abiotic stress, wounding and plant growth regulator. Sequence homology and phylogenetic analyses indicate Podhn encodes an acidic dehydrin (pI 5.14, 277 amino acids, predicted size 25.6 kDa… Show more

“…All cells and leaves were immediately frozen in liquid nitrogen and stored at − 70°C until analyzed. Isolation of total RNA and northern blot analysis were conducted as previously described by Bae et al (2009). The signal quantification was conducted using densitometric calculation by ImageJ 1.4 software (http://rsb.info.nih.gov/ij).…”

“…Expression of DHN genes can be induced by various conditions, including water deficit, salinity, low temperature (Rorat 2006), ABA, GA, SA (Sun et al 2006), JA, H 2 O 2 , wounding (Bae et al 2009), UV radiation (Allagulova et al 2003;Rorat et al 2006), heavy metals (Bae et al 2009;Zhang et al 2006), and photoperiod (Welling et al 2004). Some dehydrins can also be expressed constitutively during normal growth (Rodriguez et al 2005;Rorat et al 2004Rorat et al , 2006.…”

“…Transcriptional regulation of DHN genes in response to stress have been reported for different plant species including Capsicum annuum (Chung et al 2003), Boea crassifolia (Shen et al 2004), Oryza sativa (Lee et al 2005), Capsella bursapastoris (Fan and Wang 2006), Triticum durum (Brini et al 2007), Triticum aestivum L. (Shakirova et al 2009), Populus alba 9 P. tremula var. glandulosa (Bae et al 2009), and Prunus persica L. (Bassett et al 2009). Most responsive to abscisic acid (RAB) or cold-regulated (COR) proteins also belong to this group (Nylander et al 2001;Sun and Lin 2010).…”
