Discovering Low Toxicity Ionic Liquids for Saccharomyces cerevisiae by Using the Agar Well Diffusion Test

Abstract: Ionic liquids (ILs) are new solvents widely used in many technologies due to their unique and advantageous physicochemical properties. In biotechnological applications, ILs can be used along with microorganisms such as Saccharomyces cerevisiae. Due to the enormous number of ILs that can be synthesized through the combination of different anions and cations, it is necessary to have an easy and quick tool for the preliminary screening of their biocompatibility for being used in biotechnological applications. In … Show more

“…In addition, the most used microorganism for industrial production of ethanol, Saccharomyces cerevisiae (Meyen ex E.C. Hansen 1883), is not naturally able to ferment pentoses [8][9][10]. Among the few yeast strains that can naturally ferment xylose, Scheffersomyces stipitis ((Pignal) Kurtzman and M. Suzuki 2010), is one of the most studied, being able to reach high ethanol yields [11].…”

“…Furthermore, in order to confirm the initial screen, antibacterial activities of the isolated strains were performed by using the agar well-diffusion method [ 20 ]. Actinobacteria strains were grown for 7 days at 28 °C in the Bennet production medium.…”

“…Two to three identical colonies were picked from the plate and transferred to the broth. Each strain to be tested was grown overnight in LB media, and its OD600 was determined [ 20 ]. Seven mm (diameter) wells were perforated in the agar, and 50 μL of each Actinobacterial culture were poured into the well.…”

“…The antimicrobial activity was performed by agar well diffusion assay [37,38] . Four pathogenic microbial species including Gram‐negative Bacteria ( Klebsiella pneumonia ATCC700603, Escherichia coli ATCC25922) and Gram‐positive bacteria ( Staphylococcus aureus ATCC25923 and Streptococcus pyogenes EMCC1772) were screened.…”
