Distribution of Α‐ and Β‐adrenoceptors in Human Urinary Bladder

Abstract: The distribution of α‐ and β‐adrenoceptors in isolated preparations of human bladder neck and detrusor muscle has been studied. Adrenaline caused contraction of the bladder neck which was blocked by phenoxybenzamine but unaltered by propranolol. Isoprenaline caused relaxation of the bladder neck which was blocked by propranolol. High concentrations caused contraction which was enhanced by propranolol but blocked by phenoxybenzamine. Detrusor muscle was relaxed by isoprenaline and this effect was blocked by pro… Show more

“…The function of sympathetic innervation is opposite to that of parasympathetic innervation. The release of sympathetic neurotransmitter noradrenalin causes that the detrusor is blocked by inhibitory β-adrenergic receptors and both the bladder neck and the smooth-muscular urethra are tonicized by excitatory detrusor α-adrenergic receptors, thus achieving continence (Nergardh and Boreus 1972, Edvardsen and Setekleiv 1968, Salimi et al 1969, Raezer et al 1973, Awad et al 1974, Levin and Wein 1979. Therefore, the observed increase in the number of adrenergic nerve fibers could be a factor which additionally decreases the spasticity of the overreactive bladder and for that reason improves the treatment.…”

“…The presence of adrenergic receptors in the human lower urinary tract has been demonstrated both in the outflow region (Awad et al, 1974) and in the prostate (Caine et al, 1975). The pharmacological blockade with phentolamine of the alpha receptors causes a relaxation of the urethral smooth muscle fibres, with decrease in urethral pressure and urethral profile length (Donker et al, 1972;Whitfield et al, 1976).…”

“…Other studies are restricted to a specific connection or function of the neuronal circuitry (e.g. Andersson 1986;Awad et al 1974;Baljet and Drukker 1979;Bradley 1969a, b;Bradley and Conway 1966;Teague 1968a, b, 1969a~1;Coote et al 1982;De Groat and Saum 1971;Donker 1986;DroEs 1972;Elbadawi 1984;Gosling et al 1983;Griffiths et al 1990;Hickey et al 1982;Marani et al 1993;Raz 1978;Tanagho et al 1982;Van Ulden 1975;Venema 1988).…”
