Identification and quantification of fish diet diversity was the first step in understanding the food web dynamics and ecosystem energetics, where the contribution of DNA barcoding technique has been important. We used DNA barcoding to identify the stomach contents of a euryhaline, benthophagous catfish Ariius maculatus. From 40 catfish stomach items sampled in two different seasons, we barcoded 67 prey items of chordates and macro-invertebrates identified as belonging to 13 species in 4 major phyla (viz., Chordate, Arthropod, Annelida and Mollusca). It is important to note that the mollusc taxa (Meritrix meritrix and Perna viridis) and the species of fish (Stolephorus indicus) could not be found among the gut contents of A. maculatus sampled during the pre- and post-monsoon season, respectively. Among the chordate diets of A. maculatus, Eubleekeria splendens (23.5%) and Stolephorus indicus (23.5%) were the major prey taxa during pre-monsoon season. The hermit crabs forms the major constituents of both pre- and post-monsoon seasons, among invertebrate taxa. Polychaete, Capitella capitata (25.92%) was abundantly consumed invertebrates next to hermit crabs. We noticed that in pre-monsoon A. maculatus was more piscivorous than post-monsoon. As revealed through Kimura-2 parametric pair-wise distance analysis, the diet diversity was relatively higher in post-monsoon. The accumulation curve estimated 57 haplotypes within 14 barcoded species (including the host A. maculatus). Majority of haplotypes were found in Chordates (47.36%) followed by Arthropods (28.07%), Annelids (14.03%) and Mollusca (10.52%), respectively. This study also highlights that there is a growing concern about A. maculatus’s aggressive predation on commercially important stocks of fish and invertebrates. We will continue to expand the coverage of species barcoded in the reference database, which will become more significant as meta- and environmental DNA barcoding techniques become cheaper and prevalent.

Pengelolaan dan pelestarian keanekaragaman hayati berawal dari pengetahuan tentang ketersediaan sumberdaya, identifikasi, dan sistematika spesies. Banyaknya spesies Turbinidae dengan berbagai bentuk morfologi dan transformasi bentuk menjadikan identifikasi dengan gen COI sangat penting dalam mempercepat pengungkapan identitas spesies. Penelitian ini dilakukan dengan tujuan menganalisis pola pertumbuhan dan mengidentifikasi gastopoda Turbinidae dengan pendekatan DNA barcode. Hasil identifikasi berdasarkan karakter morfologi menunjukkan bahwa genus Turbo yang diamati adalah dari spesies Turbo stenogyrus. Analisis pola pertumbuhan menunjukkan bahwa pola pertumbuhan T. stenogyrus adalah allometrik negatif. Amplifikasi DNA dengan gen COI menghasilkan panjang sekuen DNA 650 bp dengan hasil identifikasi sesuai dengan pengamatan morfologi yaitu Turbo stenogyrus. Berdasarkan analisis filogenetik dan jarak genetik, sekuen DNA yang dianalisis membentuk clade yang sama dengan spesies Turbo stenogyrus dengan jarak genetik terdekat 4,2%. Analisis genetik melalui gen COI sangat baik dan memberi kemudahan dalam identifikasi spesies gastropoda.

Abstract. Nurilmala M, Atjitama Y, Jacob AM, Indarwati AR, Nugraha R. 2020. Advancing traceability using DNA sequence on seafood product: fraudulence in shrimp crackers. Biodiversitas 21: 5650-5656. Shrimp crackers are snacks made from starch with the addition of shrimp and other ingredients. Authentication of shrimp crackers becomes a quality control method in line with its traceability since the morphological characters are not recognized in those products. The failure of the authentication will lead to adulteration of shrimp products. The study aimed to design shrimp-specific polymerase chain reaction (PCR) primers by and used them to authenticate shrimp cracker products to assure their traceability. The primers were designed from cytochrome oxidase subunit I (COI) gene with a GC content of 50% and a melting temperature of 57 °C. The amplicon had a length of 613 bp. Fourteen shrimp crackers having DNA concentrations 0.3 to 8.1 ng/µL were evaluated. Eight out of fourteen DNA samples were successfully amplified using the specific primer and could be visualized on the agarose gel at 500-750 bp. Those DNA samples were identified belonging to shrimp species, namely Litopenaeus vannamei, Metapenaeus ensis, and Fenneropenaeus merguiensis with 95% to 100% identity. Meanwhile, the other six DNA samples underwent PCR using universal primers. One sample was amplified and identified as fish (Sphyraena flavicauda). These results indicate illegal substitution of shrimp using unidentified species occurred in shrimp crackers.
