“…The slides were post-fixed for 10 min in a solution of formaldehyde/MgCl 2 (1% formaldehyde/50 mM MgCl 2 in PBS). The slides after denaturation (2X SSC at 70°C for 20 min and after cooling to ambient temperature 1 min in 0.07 N NaOH) were dehydrated in graded series of ethanol (30,70, 90 and 100%) and air dried. The M-FISH probe mixture was denatured separately by incubation at 75°C for 5 min followed by incubation at 37°C for 30 min to allow the annealing of repetitive DNA sequences.…”
