Effect of cigarette smoke and its condensates on alveolar epithelial cell injury in vitro

Abstract: The oxidant-antioxidant balance in the airspaces of the lungs may be critical in protecting the lungs from the effects of cigarette smoke. We studied the effect of cigarette smoke and its condensates on the detachment, attachment, and proliferation of the A549 human alveolar epithelial cell line, in an in vitro model of cell injury and regeneration and the protective effects of antioxidants. Whole and vapor phase cigarette smoke decreased 51Cr-labeled A549 cell attachment, increased cell detachment, and decrea… Show more

“…CSC was produced by blowing smoke, generated by a smoking machine using a vacuum syringe system, over 3 ml of phosphate buffered saline (PBS) in a siliconised glass tonometer (Vitalograph, Buckingham, UK). The smoke-generating machine delivers a 37 ml puff of whole cigarette smoke, including particulates, every minute to the tonometer which was rotated gently [14]. The solution was made up fresh on the morning of each experiment and CSC (100%) was filtered L Rahman et aL/FEBS Letters 396 (1996) [21][22][23][24][25] using 0.22 ~tm filter (Millipore S.A., Molsheim, France) to remove large particles and bacteria.…”

“…Several studies (Fukano et al, 2006a,b;Hoshino et al, 2001;Lannan et al, 1994) have investigated the effects of smoking machine-generated CSC or whole smoke on cultured cell lines, particularly A549 cells, a transformed cell line with some characteristics of alveolar type II cells.…”
