Effect of the quaternary ammonium compound trospium chloride on 24 hour jejunal motility in healthy subjects.

Abstract: This study aimed to record 24 hour jejunal motility in healthy ambulant subjects and to analyse changes in motility caused by the oral administration of an anticholinergic agent, the quarternary ammonium compound, trospium chloride. In a placebo-controlied, double blind crossover trial, 24 hour jejunal motility was recorded in 12 healthy volunteers, aged 25 (21-30) years, using a digital data logger connected to two strain-gauge transducers mounted 20 cm apart in a flexible nasojejunal catheter. A computer pro… Show more

“…During the interdigestive period there were three quantitative parameters that were significantly different between the patient group and the healthy controls: in dyspeptics the incidence of antral or jejunal phase 3s was reduced and the relative contribution of phase 2 to the MMC cycle was increased. These observations confirm previous findings,6 7 and the reference values obtained in the healthy volunteers are in accordance with other studies using ambulatory manometry 2425 Phase 3 has been interpreted as having a housekeeper function by causing rapid aboral and effective propagation of intraluminal contents 19.…”

“…The chemical structures of oxybutynin, tolterodine, and trospium chloride are depicted in Figure 1 [6,7]. The major difference between tertiary and quaternary amines is the (+) charge and hydrophilic nature of trospium chloride, a quaternary amine.…”

“…Intubation of the small intestine was performed as described in detail previously [5]. Briefly, after an overnight fast, the subjects were intubated transnasally with the MII manometry system after local anesthesia of nose and throat (Xylocaine Spray; Astra Chemicals, Wedel/Holstein, Germany).…”

“…Manometry has established as a tool in clinical practice to test the integrity of the neuromuscular apparatus of the small intestine [2,3,4,5]. However, manometric studies do not provide information on the transport processes in the intestinal lumen.…”
