Effective inhibition of Th17/Th22 pathway in 2D and 3D human models of psoriasis by Celastrol enriched plant cell culture extract

Abstract: BackgroundPsoriasis is an immune‐mediated inflammatory disease in which the Th17 pathway is mainly involved. Systemic interventions with biologics that specifically block the Th17 pathway are effective to treat severe psoriasis. However, for efficient topical treatment, small molecules are more suitable than antibodies to penetrate and target epidermal keratinocytes, the key players in psoriasis. Celastrol, a well‐described triterpene, is present in low amounts in Tripterygium wilfordii roots. By using plant c… Show more

“…The mechanism thought to be involved in the alleviation of multiple sclerosis by Celastrol includes the modulation of Th17 responses. CEE has been tested in 2D (human CD4+ T cells and normal human epidermal keratinocytes) and 3D RHE in vitro assays to determine its impact on markers relating to psoriasis 36 . In these studies, Th17/Th22 cytokines were inhibited by CEE in both 2D and 3D models.…”

“…Furthermore, celastrol downregulated the expression of cytokines and chemokines, such as IL-8, IL-19, IL-23, CCL5, CCL20, and CXCL1, and inhibited the T cell differentiation from Th17 and Th22 in a 2D and 3D human in vitro model of psoriasis. [ 115 ]. There are no clinical trials in humans with autoimmune diseases.…”

“…Multi-glycoside of Tripterygium wilfordii Hook. f (GTW), also termed as Tripterygium glycosides, is a widely acknowledged efficacious psoriasis-treating TCM agent and has been proven to ameliorate murine psoriasis-like dermatitis, which was therefore chosen as positive drug in our study (Han et al, 2012;Wu et al, 2015a;Zhao et al, 2016;Lv et al, 2018;Nguyen et al, 2020a;Ru et al, 2020). As modeling with IMQ generated skin lesions resembling human plaque psoriasis, mice pretreated with XYF displayed lighter erythema, smoother skin, and thinner scales than mice treated with IMQ alone (Figure 2B).…”
