Effects of Locally Administered Tiludronic Acid on Experimental Periodontitis in Rats

Abstract: It can be concluded that locally administered TIL solution (1 mg/kg body weight) reduced alveolar bone loss in experimental periodontitis and the dosage of TIL may influence its anti-inflammatory and antiresorptive properties.

“…For volumetric measurements, a volume of interest (VOI‐prismatic section) was outlined from the apexes of all roots of M1 up to the roof of the furcation of M1, touching the roots surfaces, 21 in all images of the coronal dataset, using appropriate software ****. The following parameters were analyzed: 1) BV: percentage of VOI filled with bone tissue; 2) bone porosity (BP): percentage of porosities present in the bone tissue within the VOI; 3) trabecular number (Tb.N): number (mm ‐1 ) of bone trabeculae present within the VOI; 4) trabecular separation (Tb.Sp): total of spaces (mm) present among bone trabeculae within the VOI.…”

“…Immunohistochemical processing was performed through the indirect immunoperoxidase method, as described elsewhere [21]. The histological slides containing samples from all animals were incubated with one of the following primary antibodies: rabbit anti-interleukin (IL)-1β (1:100-Rabbit anti-IL-1β–SC 7884, Santa Cruz Biotechnology, Santa Cruz, CA, USA), goat anti-cytokine-induced neutrophil chemoattractant–CINC (1:100-Goat anti-CINC1–ab 10365, abcam ® , Cambridge, MA, USA), goat anti-IL-10 (1:100-Goat anti-IL-10–SC 1783, Santa Cruz Biotechnology), rabbit anti-transforming growth factor (TGF)-β1 (1:200-Rabbit anti-TGF-β1–SC 146, Santa Cruz Biotechnology) or goat anti-tartrate-resistant acid phosphatase–TRAP (1:100-Goat anti-TRAP–SC 30833, Santa Cruz Biotechnology).…”

“…Immunohistochemical processing was performed through the indirect immunoperoxidase method, as described previously . Histologic slides containing samples from all the experimental groups were divided into three batches, and each batch was incubated with one of the following primary antibodies: 1) goat anti‐tartrate‐resistant acid phosphatase (TRAP); 2) goat anti–receptor activator of nuclear factor‐κB ligand (RANKL); 3) goat antiosteoprotegerin (OPG); or 4) rabbit anti–cleaved caspase 3 (CC3)…”

“…A quantitative immunolabeling analysis was performed for TRAP and CC3. TRAP‐positive cells were distinguished as TRAP‐positive multinucleated cells (mature osteoclasts) or TRAP‐positive mononuclear cells . Only osteocytes CC3‐positive were quantified.…”
