Efficacy of a Swab Transport System in Maintaining Viability of Neisseria gonorrhoeae and Streptococcus pneumoniae

Abstract: The efficacy of swab transport systems in maintaining viability of Neisseria gonorrhoeae and Streptococcus pneumoniae is crucial both for establishing definitive diagnosis and for monitoring emerging resistance. We tested the efficacy of a newly modified Amies charcoal swab transport system, the StarSwab SP131X (Starplex Scientific, Inc., Etobicoke, Ontario, Canada), by using a combined total of 31 clinical and American Type Culture Collection stock reference strains of N. gonorrhoeae and S. pneumoniae in 46 s… Show more

“…Our observations may explain variations in viability among strains of N. gonorrhoeae described by others (3,7,8) and suggest that in vitro studies using broth suspensions to compare plain and charcoal media may have underestimated the performance of plain media (1).…”

“…In vitro and clinical evaluations of transport medium for Neisseria gonorrhoeae reveal variation in survival of uncharacterized isolates within and between such studies (3,7,8). In the present study, we investigated whether auxotypes of N. gonorrhoeae differ in their abilities to survive in plain and charcoalcontaining transport media.…”

“…The effect of pooling samples, collected as swabs or organ samples, on the detection of TSRV by RT-qPCR was investigated. While the use of swabs has been described for collecting samples to detect viruses by PCR for humans and terrestrial animals (Mahony & Chernesky 1985;Roelofsen et al 1999;Farhat, Thibault & Devlin 2001;Cloud, Hymas & Carroll 2002;Jordens et al 2002;Echavarria et al 2003;Uhl et al 2003;Issa et al 2005), this approach has not been evaluated for the detection of fish viruses by PCR.…”

“…In: Proceedings of the 104th general meeting of the American Society for Microbiology, New Orleans, LA). 9,12,19 Scoring was as follows: 0 5 no growth; 1 5 growth of 1-20 colonies in the first quadrant; 2 5 growth of .20 colonies in the first quadrant; 3 5 growth in the first and second quadrants; 4 5 growth in the first, second, and third quadrants; and, 5 5 growth in all 4 quadrants. Differences in growth scores between baseline (i.e., swabs plated immediately after inoculation) and the 2 storage conditions (room temperature for 1 hr and 280uC for 40 days) were analyzed using Friedman repeated measures analysis of variance on ranks; if a significant difference was found, the individual storage conditions were compared with the baseline using the Holm-Sidak method for pairwise comparisons.…”
