Efficacy of Alpinumisoflavone Isolated from Maclura tricuspidata Fruit in Tumor Necrosis Factor-α-Induced Damage of Human Dermal Fibroblasts

Abstract: The skin is an important organ in the human body that protects the body from environmentally hazardous substances. Reactive oxygen species (ROS) cause inflammatory reactions and degradation of the extracellular matrix leading to skin aging and various cutaneous lesions. This study evaluated the potential of isoflavones isolated from Maclura tricuspidata fruit to prevent TNF-α-induced skin inflammation in normal human dermal fibroblasts (HDFs). It focused on alpinumisoflavone (AIF) that suppressed the accumulat… Show more

“…A tumor necrosis factor-α (TNF-α)-induced damaged fibroblast could also be alleviated by several polyphenols [ 48 , 49 , 50 , 51 ] ( Table 1 ). Alpinumisoflavone (AIF), (-)-catechin, epigallocatechin-3-gallate (EGCG), and 7,8-dihydroxyflavone (7,8-DHF) suppressed the TNF-α-induced MMP-1 synthesis and enhanced procollagen I [ 48 , 49 , 50 , 51 ]. AIF and (-)-catechin inhibited NF-κB activity and COX-2 [ 48 , 49 ].…”

“…The reaction was performed using the QuantStudio™ 3 Real-Time PCR System (Applied Biosystems), and the thermal conditions were set to 40 cycles (95 °C for 15 s, 60 °C for 30 s, 95 °C for 30 s). The primer sequences are shown in a previous paper [ 40 ], and β-actin was used as the reference gene.…”

“…Change of photoaged skin is closely related to degradation of collagen and elastic fibers in the dermis, [29] which is featured by sagging, wrinkling, coarseness, and erythema [30] . Photoaging stimulates the expression of intracellular ROS, which is a major part of the inflammatory response and ECM degradation that in turn leads to skin aging and various cutaneous lesions [31] …”

“…[30] Photoaging stimulates the expression of intracellular ROS, which is a major part of the inflammatory response and ECM degradation that in turn leads to skin aging and various cutaneous lesions. [31] The representative photographs of the UV-irradiated skin lesions at the ninth week were presented in Figure 2A, including skin appearance and skin pinch testing. Throughout the entire experiment, the dorsal skin in the SC group was healthy, suggesting that the shaving approach did not influence the skin morphology.…”
