Efficiency of chitinolytic enzymes in the formation of Tricholoma matsutake protoplasts.

Abstract: Since the cell walls of basidiomycetes consist of chitin and other things,1} chitinolytic enzymes seem to be essential for protoplast formation. There are considerable numbers of reports on the formation of basidiomycete protoplasts, but only one on Tricholoma matsutake.2) Chitinase was effective for protoplast formation from Lentinus edodes^and other basidiomycetes in the presence of Cellulase Onozuka RS. Furthermore, Trichoderma harzianum isolated from L. edodes logs produced chitinase and /?-l,3-glucanase, … Show more

“…The β-1,3-glucanase activity in seed proteins was assayed by measuring the rate of reducing sugar (glucose) generated using laminarin as a substrate (Koga et al 1988). Briefly, the reaction mixture consisted of 20 μl laminarin (1%) and various volumes (2.5, 10 μl) of seed protein in the assay buffer of 0.05 M sodium acetate pH 5.2 (100 µl).…”

“…β-1,3-glucanase activity was estimated using the procedure of Koga et al [22]. The assay mixture contained 1.0 ml of suitably diluted enzyme and 1.0 ml of 1% laminarin solution in sodium acetate buffer (50 mM, pH 5.2).…”
