Efficient production of gamma‐aminobutyric acid by engineered Saccharomyces cerevisiae with glutamate decarboxylases from Streptomyces

Abstract: Gamma‐aminobutyric acid (GABA) is an industrially valuable natural product. This study was aimed to establish an efficient food‐grade production process of GABA by engineering Saccharomyces cerevisiae that is generally recognized as safe (GRAS). GABA can be produced by catalytic decarboxylation of l‐glutamate (l‐Glu) by glutamate decarboxylase (GAD, EC4.1.1.15). Two GADs, SsGAD from Streptomyces sp. MJ654‐NF4 and ScGAD from Streptomyces chromofuscus ATCC 49982, were heterologously expressed in S. cerevisiae BJ… Show more

“…When engineered E. coli was used as the biocatalyst, the production or productivity was the highest among all the whole-cell bioconversion catalysts thus far. For example, by considerable modification of a recombinant GABA-producing E. coli strain, 308.3 g/L GABA was produced within a single whole-cell conversion process with 44.04 g/L/h productivity and 99.6 mol % conversion in a 5 L bioreactor .…”

“…Similarly, Yuan et al. reported the production of 62.6 g L −1 (60.8 %) GABA from 0.1 M glutamate by engineering Saccharomyces cerevisiae expressing GAD from Streptomyces 99. Yuan also reported similar work on an engineered E .coli strain which expressed GAD from Streptomyces and produced and 2771 g L −1 (67 %) GABA 100.…”

“…Engineered S. cerevisiae was successfully used to produce a high level of natural products from other sources, for example, artemisinic acid up to 100 mg/L [71], tetrahydrocannabinolic acid up to 8.0 mg/L, cannabidiolic acid, and tetrahydrocannabivarinic acid up to 4.8 mg/L [72]. Engineered S. cerevisiae also produced some natural products from Streptomyces with high titers, such as gamma aminobutyric acid up to 62.6 g/L [73] and cannabigerolic acid up to 299.8 mg/L [74].…”
