Elucidation of the Mass Fragmentation Pathways of Tomatidine and β1-Hydroxytomatine using Orbitrap Mass Spectrometry

Abstract: Tomatoes, members of the Solanaceae plant family, produce biologically active secondary metabolites, including glycoalkaloids, which may have both adverse and beneficial biological effects. Using the linear ion trap (LIT) mass spectrometry, multi-stage collision induced dissociation (CID) experiments (MSn) were performed to elucidate characteristic fragmentation pathways of the glycoalkaloid, tomatidine and of β1-hydroxytomatine. High resolution with high accuracy mass analysis using an Orbitrap fourier transf… Show more

“…As with tomatidine, these metabolites were also detected in the tangerine and low carotenoid groups, providing added evidence of them as biomarkers of tomato intake. Their fragmentation patterns are similar to tomatidine and can be rationalized based on the MS/MS fragment structures published by Caprioli et al . All metabolites appear to have a 6 ring structure with a steroidal backbone and fused aminoketal end rings.…”

“…However, to the best of our knowledge, there have been no studies of the fragmentation of bisphenols based on high‐resolution (HR)MS data, which would enable a more detailed and systematic investigation of the fragmentation pathways of bisphenols to be performed. Orbitrap MS is a powerful tool for characterizing trace amounts of compounds by establishing fragmentation pathways . Accurate mass information is important for the confirmation of the identities of product ions and plays a key role in the elucidation of fragmentation pathways.…”

“…Orbitrap MS is a powerful tool for characterizing trace amounts of compounds by establishing fragmentation pathways. [23][24][25] Accurate mass information is important for the confirmation of the identities of product ions and plays a key role in the elucidation of fragmentation pathways. Isotope labeling is also a good method for investigating fragmentation pathways, [26][27][28] with carbon-13 ( 13 C) [29] and deuterium (D) [30] often being used as labeling isotopes.…”

“…Tomatidine is a steroidal alkaloid that has been found in the skins and leaves of tomatoes [93]. Different therapeutic effects have been described for tomatidine such as antiasthmatic [94], anti-inflammatory [95], antimicrobial [96,97], and anticancer [98] effects.…”
