Endocytosis of interleukin 2 receptors in human T lymphocytes: distinct intracellular localization and fate of the receptor alpha, beta, and gamma chains.

Abstract: Abstract. Members of the cytokine receptor family are composed of several noncovalently linked chains with sequence and structure homologies in their extracellular domain. Receptor subfamily members share at least one component: thus the receptors for interleukin (IL) 2 and ILl5 have common ~ and 3" chains, while those for IL2, 4, 7, and 9 have a common 3' chain. The intracellular pathway followed by IL2 receptors after ligand binding and endocytosis was analyzed by immunofluorescence and confocal microscopy i… Show more

“…All three subunits initially colocalize with Tfn-R-loaded endosomes. Whereas the ␤ and ␥ chains are subsequently routed to rab7-positive late endosomes for degradation, the ␣ chain is recycled back to the cell surface via recycling endosomes (Hemar et al, 1995). CCR5 could be recycled in an analogous manner.…”

“…Several members of the Type I cytokine receptor family have been shown to undergo ligandinduced ubiquitination and internalization [17,[20][21][22][23][24][25]. Perhaps the best characterized of these is the GHR.…”

“…For the Type I cytokine receptors, endocytosis and degradation has been best characterized for the growth hormone receptor (GH-R), interleukin-2 receptor (IL-2R), and the erythropoietin receptor (EpoR) [20][21][22][23][24][25]. These receptors all undergo ligand-induced ubiquitination.…”

“…This observation means that there really was less CD25 on the surface of the IL-2 cultured cells because this particular anti-CD25 monoclonal antibody binds to IL-2R alpha even when IL-2 is already bound to the receptor [36]. In the presence of IL-2, the CD25 IL-2R alpha receptor may be cycling, removing the CD25 protein from the surface of the cell [37]. As expected, the transferrin receptor CD71 was up regulated (Fig.…”
