Engineered stem cells targeting multiple cell surface receptors in tumors

Abstract: Multiple stem cell types exhibit inherent tropism for cancer, and engineered stem cells have been used as therapeutic agents to specifically target cancer cells.Recently, stem cells have been engineered to target multiple surface receptors on tumor cells, as well as endothelial and immune cells in the tumor microenvironment. In this review, we discuss the rationales and strategies for developing multiple receptor-targeted stem cells, their mechanisms of action, and the promises and challenges they hold as cance… Show more

“…To address these problems, MSCs have been transplanted in vivo combined with scaffolds, cultured as a sheet form, or engineered to have targeting ligands. [61][62][63] Likewise, it has been known that spheroids could enhance the engraftment efficiency within the injected area due to their relatively larger volume compared to suspended cells, which increase the therapeutic efficacy of transplanted MSCs. For example, MSC spheroids injected into an ischemic kidney showed an enhanced survival rate and paracrine signaling as compared to dissociated cells by increased retention of transplanted cells within the transplanted tissue.…”

“…Additionally, MSCs as a possible means can deliver oncolytic viruses (OVs), which can overcome host antiviral immunity [ 159 ]. MSCs armed by oncolytic herpes simplex virus (oHSV) successfully delivered viral progeny to target melanoma brain metastasis, ultimately inhibiting tumor growth rate and prolonging survival of brain tumor-bearing mice [ 159 , 160 ]. Although many research examples of immunomodulation and delivery are successful, we still need more work.…”

“…They play the central role in the homeostasis and regeneration of all body tissues. SCs exhibit inherent tropism for cancer cells, which makes them an ideal therapeutic vehicle in anticancer therapy [105,106] (Table 2).…”
