Engineering of Adipose Tissue by Injection of Human Preadipocytes in Fibrin

Abstract: The findings demonstrate that long-term stable adipose tissue can be engineered in vivo by simple injection of human preadipocytes using fibrin as a carrier material. After further investigation, this approach may represent an alternative to the techniques currently used for soft tissue restoration.

“…Adipogenic differentiation did not significantly affect the hLCAT delivery and the cell survival in this model using fibrin glue as a scaffold with m-ccdPA ( Figure 2B). In this context, our results may suggest that the transplanted cells with fibrin glue were differentiated into adipocytes without adipogenic pretreatment (Cho et al, 2006;Torio-Padron et al, 2007a). The immunohistochemical observation did not clearly show that ccdPA would undergo adipogenic differentiation after transplantation, but the transplanted ccdPA were clearly identified as hLCAT-delivery cells in the transplanted sites of the recipient mice ( Figures 3A-3F).…”

“…Im Falle eines ausgedehnten Defektes im Gesichtsbereich bietet sich die allogene Gesichtstransplantation als potenzielle operative Lösung an, nicht zuletzt weil das "tissue engineering" [23] und die Stammzellenforschung [13] bisher keine echte klinische Alternative bieten.…”

“…Isolation of pre-adipocytes was performed directly after harvesting of the materials as described before [Torio-Padron et al, 2007]. To isolate pre-adipocytes from excised tissue, fibrous structures and visible blood vessels were removed, tissue was minced into small pieces using a scalpel and washed in PBS.…”

“…Isolated cells were differentiated into adipocytes using a differentiation medium based on Dulbecco's modified Eagle's medium/Ham's F-12 medium 1: 2 [Torio- Padron et al, 2007]. This medium was mixed with 3% fetal bovine serum, insulin 100 n M (Sigma, Deisenhofen, Germany), dexamethasone 1 M (Sigma), and, for the first 4 days of differentiation, methylisobutylxanthine 0.2 m M (Sigma).…”
