Enzyme Regulation in C4 Photosynthesis

Abstract: Activities catalyzing the synthesis of fructose-2,6-bisphosphate (fructose-6-phosphate,2-kinase or Fru-6-P,2K) and its breakdown (fructose-2,6-bisphosphatase or Fru-2,6-P2ase) were identified in leaves of corn (Zea mays), a C4 plant. Fru-6-P,2K and Fru-2,6-P2ase were both localized mainly, if not entirely, in the leaf mesophyll cells. A partially purified preparation containing the two activities revealed that the kinase and phosphatase were regulated by metabolite effectors in a manner generally similar to th… Show more

“…For maize leaves, evidence has also been obtained for the primary localization of 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase (27) and cytoplasmic fructose-1 ,6-bisphosphatase (5) activities in the MC. However, a strict compartmentation (as in the case for carboxylating enzymes) has not been established for any of these enzymes and it is certainly possible that 10 to 40% of the whole leaf activities could be contained in the BSC.…”

“…The biphasic substrate saturation curve reported by several groups working with plants (4,19) can probably be attributed to the use of a mixture of the low-and high-Km enzymes. The respective role of the two types of enzyme in the hydrolysis of Fru-2,6-P2 in vivo has not been defined.…”

“…13 as C4 plants in which there is a spatial separation between photosynthetic CO2 fixation and sucrose synthesis (7). In addition, the Fru 6-P,2K and Fru 2,6-P2ase from a C4 plant have been found to differ in certain regulatory characteristics from their C3 counterparts (15 1.5% (w/v) insoluble polyvinyl polypyrrolidone. Rats were killed by decapitation, the livers excised immediately, and 20 g of the tissue was extracted as described for leaves.…”
