Mid-infrared (MIR) spectroscopy is the method of choice for the standard milk recording system, to determine milk components including fat, protein, lactose and urea. Since milk composition is related to health and metabolic status of a cow, MIR spectra could be potentially used for disease detection. In dairy production, mastitis is one of the most prevalent diseases. The aim of this study was to develop a calibration equation to predict mastitis events from routinely recorded MIR spectra data. A further aim was to evaluate the use of test day somatic cell score (SCS) as covariate on the accuracy of the prediction model. The data for this study is from the Austrian milk recording system and its health monitoring system (GMON). Test day data including MIR spectra data was merged with diagnosis data of Fleckvieh, Brown Swiss and Holstein Friesian cows. As prediction variables, MIR absorbance data after first derivatives and selection of wavenumbers, corrected for days in milk, were used. The data set contained roughly 600,000 records and was split into calibration and validation sets by farm. Calibration sets were made to be balanced (as many healthy as mastitis cases), while the validation set was kept large and realistic. Prediction was done with Partial Least Squares Discriminant Analysis, key indicators of model fit were sensitivity and specificity. Results were extracted for association between spectra and diagnosis with different time windows (days between diagnosis and test days) in validation. The comparison of different sets of predictor variables (MIR, SCS, MIR + SCS) showed an advantage in prediction for MIR + SCS. For this prediction model, specificity was 0.79 and sensitivity was 0.68 in time window -7 to +7 days (calibration and validation). Corresponding values for MIR were 0.71 and 0.61, for SCS they were 0.81 and 0.62. In general, prediction of mastitis performed better with a shorter distance between test day and mastitis event, yet even for time windows of -21 to +21 days, prediction accuracies were still reasonable, with sensitivities ranging from 0.50 to 0.57 and specificities remaining unchanged (0.71 to 0.85). Additional research to further improve prediction equation, and studies on genetic correlations among clinical mastitis, SCS and MIR predicted mastitis are planned.

ABSTRAK. Kesehatan ambing merupakan faktor penting dalam usaha peternakan sapi perah. Kerugian yang ditimbulkan ketika kesehatan ambing tidak terjaga adalah munculnya kasus mastitis atau sering disebut dengan mastitis. Ternak yang terinfeksi mastitis akan mengalami penurunan kualitas dan kuantitas susu sehingga berdampak pula terhadap pendapatan ekonomi peternak. Kasus mastitis yang sering terjadi pada ternak adalah mastitis subklinis. Mastitis subklinis merupakan radang pada ambing yang terjadi tanpa disertai kemunculan gejala klinis namun kualitas dan kuantitas susu yang dihasilkan menurun secara drastis. Tingginya angka kejadian mastitis subklinis mendorong untuk dilakukan penyuluhan mengenai pengelolaan sistim pemeliharaan dan kesehatan pada sapi perah sebagai upaaya pencegahan kasus mastitis.Metode pengukuran keberhasilan penyuluhan melalui penghitungan statistik persentase hasil quisioner yang dilakukan sebelum dan sesudah penyuluhan. Topik quisioner yang diambil adalah pemahaman masyarakat terhadap kasus mastitis serta faktor -faktor yang mempengaruhi kesehatan ambing. Rangkaian kegiatan penyuluhan ini sebagai upaya untuk menekan angka kejadian mastitis melalui peningkatan pemahaman masyarakat. Hasil dari penyuluhan ini adalah peningkatan pengetahuan peternak sebesar 20,8% mengenai definisi, gejala klinis serta faktor-faktor resiko yang dapat menimbukan mastitis Kata kunci: mastitis; kesehatan; sapi perah; sumedang; pemeliharaan; penyuluhan ABSTRACT. Udder health is an important factor in dairy farming business. Losses caused when udder health is not maintained is the emergence of cases of mastitis or often referred to as mastitis. Livestock infected with mastitis will experience a decrease in the quality and quantity of milk so that it also affects the economic income of farmers. Cases of mastitis that often occur in cattle is subclinical mastitis. Subclinical mastitis is an inflammation of the udder that occurs without the appearance of clinical symptoms but the quality and quantity of milk produced drastically decreases. The high incidence of subclinical mastitis encourages counseling regarding the management of the maintenance and health system in dairy cows as an effort to prevent mastitis cases. The method of measuring the success of counseling through statistical calculation of the percentage of the results of the questionnaire conducted before and after counseling. The questionnaire topic taken was people's understanding of mastitis cases as well as factors affecting udder health. The series of outreach activities as an effort to reduce the number of mastitis events through increased public understanding. The result of this counseling was an increased farmer's knowledge by 20.8% regarding the definition, clinical symptoms and risk factors that can caused mastitis

This work aimed to evaluate the effects of CasuL on growth and viability of 15 mastitis isolates from cows and goats, to determine the synergistic potential between CasuL and antibiotics, and to investigate the effects on bacterial ultrastructure and antibiofilm activity. The lectin inhibited the growth of Staphylococcus isolates from either bovine (Ssp6PD and Sa) or caprine (Ssp5D and Ssp01) mastitis. The minimal inhibitory concentrations were ranged from 3.75 to 15 µg/ml. Synergistic effect was observed for CasuL‐tetracycline against Sa and Ssp6PD and CasuL‐ampicillin against Ssp01. No structural damage was observed under the scanning electron microscope in CasuL treatments. Flow cytometry analysis using thiazol orange and propidium iodide demonstrated that CasuL was unable to reduce the cell viability of the isolates tested. At sub‐inhibitory concentrations, CasuL reduced biofilm formation by the isolates Sa and Ssp5D. However, CasuL‐tetracycline and CasuL‐ampicillin combinations inhibited biofilm formation by Ssp6PD and Ssp01, respectively. In conclusion, CasuL is a bacteriostatic and antibiofilm agent against some mastitis isolates and displayed a synergistic potential when used in combination with either ampicillin (against one isolate) or tetracycline (against two isolates). The results stimulate the evaluation of CasuL for the treatment of mastitis, particularly when used in conjunction with antibiotics.
