Estrogenic Compounds, Estrogen Receptors and Vascular Cell Signaling in the Aging Blood Vessels

Abstract: The cardiovascular benefits of menopausal hormone therapy (MHT) remain controversial. The earlier clinical observations that cardiovascular disease (CVD) was less common in MHT users compared to non-users suggested cardiovascular benefits of MHT. Also, experimental studies have identified estrogen receptors ERα, ERβ and GPR30, which mediate genomic or non-genomic effects in vascular endothelium, smooth muscle, and extracellular matrix (ECM). However, data from randomized clinical trials (RCTs), most notably th… Show more

“…Many of the vascular effects of E 2 are mediated via estrogen receptors (ERs) (78). ERs have been identified in the female reproductive tract, mammary glands, and blood vessels of humans and experimental animals (9,47,68,76,78,93).…”

“…During menopause, herbal treatment is preferred instead of hormone therapy, since hormone replacement therapy possesses breast and uterine cancer risk (Davis 2001;Djuric et al, 2001;Lu et al, 2001;Ganry, 2002;van de Weijer & Barentsen, 2002;Mishra et al, 2003;Arjmandi et al, 2003¸ Magee & Rowland, 2004Beck et al, 2005;Howes et al, 2006). Although, primary effects are seen on pre and post-menopausal symptoms, it is also used for prevention of cardiovascular disease and osteoporosis, and regulation of blood lipid profile (Lissin & Cooke, 2000;Cesar et al, 2006;Smiley & Khalil, 2009). In diploid T. pratense callus and suspension culture, plant regeneration attempts have been investigated by various researchers (Phillips & Collins, 1979;Wang & Holl, 1988;Kasparova et al, 2006).…”

“…Aging is another important consideration in the analysis of ER function. A decline in endogenous estrogens in combination with aging may lead to alterations in ER expression or signaling and promote deleterious cardiovascular outcomes in response to hormone replacement (35). Indeed, this differential response to estrogens is present in the mRen2 model; estrogen depletion via ovariectomy is detrimental in young females but beneficial in aged females fed a high-salt diet (40).…”
