Evaluation of a Modified Cefsulodin-Irgasan-Novobiocin Agar for Isolation of Yersinia spp

Abstract: Y. enterocolitica and Y. pseudotuberculosis are important food borne pathogens. However, the presence of competitive microbiota makes the isolation of Y. enterocolitica and Y. pseudotuberculosis from naturally contaminated foods difficult. We attempted to evaluate the performance of a modified Cefsulodin-Irgasan-Novobiocin (CIN) agar in the differentiation of Y. enterocolitica from non-Yersinia species, particularly the natural intestinal microbiota. The modified CIN enabled the growth of Y. enterocolitica col… Show more

“…Furthermore, pathogenic strains sometimes grow as very small colonies missing the typical red centre on the CIN plates (18,42). Chromogenic-based agar plates have become popular in recent years for isolation and identification of Y. enterocolitica belonging to pathogenic biotypes (42).…”

“…Despite numerous reports on the occurrence of Y. enterocolitica among humans, pigs or wild boars, the detection and identification are still difficult due to an overgrowth by bacteria from poly-contaminated samples, unusual growth characteristics of Y. enterocolitica and arduous discrimination of its colonies from other bacteria exhibiting Yersinia -like colonies on cefsulodin-irgasan-novobiocin Agar (CIN) such as Citrobacter freundii , C. braakii , Aeromonas hydrophila, Enterobacter cloacae , Providencia rettgeri , and Morganella morganii [ 10 12 ]. The recommended isolation of Y. enterocolitica is carried out by using ISO 10273–2003 method [ 13 ].…”

“…To isolate presumptive strains of Y. enterocolitica, the ITC broth and CIN agar media are recommended [ 3 , 4 ]. CIN agar was devised by Schiemann in 1979 and is still useful in Y. enterocolitica isolation [ 10 12 , 19 21 ]. Morphology of Yersinia sp.…”

“…Y. pseudotuberculosis infection in humans was more likely to be severe than Y. enterocolitica infection 12) . However, it is also more difficult to isolate Y. pseudotuberculosis on CIN because of the presence of cefsulodin and inability to distinguish other mannitol-fermenting bacteria 6), 13) . In this study, all Y. pseudotuberculosis strains were able to grow on CIN with/without supplements but grew on CAYe without supplements.…”
