Evaluation of Clonal Uniformity in Six Superior Cacao Clones Based on SSR Marker

Abstract:
Propagation of cacao plants is generally carried out vegetatively. Therefore, plants that are clonally propagated should be genetically uniform. Genetic uniformity in cacao clones is also very important information for germplasm conservation and in obtaining pure parental crosses. Evaluation of genetic uniformity can be seen through analysis using SSR markers. This study aimed to determine the genetic uniformity in six cacao clones using SSR markers. This experiment was conducted at IIBCRI I… Show more

“…Genetic impurity and mislabelling are issues in massive seedling production (Pancaningtyas & Susilo, 2022). High-yielding cocoa clones used as mother trees in plant propagation are required to maintain their genetic purity (genetic homogeneity) and clone characteristics (Cruz-Martinez et al, 2017;Sulistiyorini et al, 2018). Molecular markers can be used to identify genetic stability in plants since they are stable, located in all regions of the plant, and are not influenced by the environment (Sulistiyorini et al, 2018).…”

“…Simple sequence repeat markers are developed by amplifying the targeted SSR sequences using PCR. Copyright © 2019 Universitas Brawijaya Many studies have shown the presence of a high allelic variation among different plant populations (Illahi, Wiendi, & Sudarsono, 2016;Larekeng, Purwito, Mattjik, & Sudarsono, 2018;Oktavia, Kuswanhadi, Dinarty, Widodo, & Sudarsono, 2017;Purwoko, Cartealy, Tajuddin, Dinarti, & Sudarsono, 2019;Sulistiyorini, Rubiyo, & Sudarsono, 2018). The SSR markers are used as powerful tools for supporting plant breeding of many perennial crops, including oil palm (Ajambang, Sudarsono, Asmono, & Toruan, 2012;Budiman, Apriyanto, Pancoro, & Sudarsono, 2019;Larekeng, Maskromo, Purwito, Matjik, & Sudarsono, 2015;Maskromo, Larekeng, Novarianto, & Sudarsono, 2017;Natawijaya et al, 2019;Tinche, Asmono, Dinarti, & Sudarsono, 2014).…”
