Ex Vivo Stimulation of Tumor-Draining Lymph Node Cells from Lung Cancer Patients: A Potential Resource for Adoptive Immunotherapy

Abstract: ). In our study, we established a system that T cells and DCs were stimulated together ex vivo, which was easy to conduct and produce promising results. It provided a new method for improving TDLN cell antitumor activity which might be used in the clinical cancer therapy. Cellular & Molecular Immunology. 2008;5(4):307-313.

“…Additionally, tumor‐draining lymph nodes (TDLNs) are the first immune‐inducing sites receiving tumor antigens. [ 31 ] To elucidate the adaptive immune response induced by the treatment, TDLNs from tumor‐bearing mice were collected 7 days post treatment ( Figure A). The maturation of DCs in TDLNs was analyzed by flow cytometry of the expression of CD86 and MHC‐II.…”

“…TDLNs are a good source for ex vivo expansion of immune cells. 40 41 Tumor-reactive T-cells in non-metastatic LNs can be expanded in vitro, 1 indicating their potential for ACT. Lymphocytes in SLNs are the first LNs to receive tumor antigen stimulation and possess potent tumor recognition-specific immune memory and are considered an ideal starting source of cells for ACT.…”
