Exosome‑mediated gefitinib resistance in lung cancer HCC827 cells via delivery of miR‑21

Abstract: Abstract. Acquired resistance to gefitinib remains a major challenge in cancer treatment. In the present study, the effect of exosomes on the transmission of gefitinib resistance from gefitinib-resistant HCC827 lung cancer cells (H827R) to their gefitinib-sensitive counterparts and the potential underlying mechanisms by which this occurs was investigated. Exosomes were obtained from the cell supernatant using ultracentrifugation and the ExoQuick-TC exosome precipitation solution. Drug resistance was assessed b… Show more

“…Knockdown of TGF- β 1 expression in human umbilical cord mesenchymal stem cells reverts their exosome-mediated EMT-promoting effect on lung cancer cells [ 47 ]. Exosomal contents were reported to be involved in anticancer drug resistance [ 48 ]. In addition, exosomes are involved in the crosstalk between lung cancer stem cells (CSC) and cancer-associated fibroblasts (CAF) [ 49 ], which is the primary component of the lung cancer microenvironment.…”

“…Interactions between TCs and adjacent stromal tissue may be mediated via diffusion of exosomal miRNAs 54–56. Indeed, high levels of miR-21 have been found in exosomes derived from NSCLC cell lines and exosome-mediated gefitinib resistance has been shown to be delivered via exosomal miR-21 in a LC cell line 57–59. Furthermore, lack of syndecan-1 impacts the miRNA carried in exosomes in cell-based and animal models of LC60 and plasma exosomal miR-21–5 p is among a panel of promising non-invasive prognostic biomarkers of NSCLC 61 62…”
