Exosomes derived from exhausted CD8+ T cells impaired the anticancer function of normal CD8+ T cells

Abstract: The exosomes derived from exhausted CD8+ T cells could be uptake by non-exhausted CD8+ T cells and subsequently impaired the function of receipt cells. Exosomes secreted from exhausted CD8+ T cells have distinct lncRNA expression profiles which are significantly different from those in exosomes secreted by non-exhausted CD8+ T cells.

“…Meanwhile, the exhausted CD8 + T cells, which express TIM-3 and PD-1 during the antitumor immune responses, exhibit reduced proliferation and impaired anticancer activities. The regular CD8 + T cells uptake exosomes containing lncRNAs secreted by the exhausted CD8 + T cells, which leads to CD8 + T cell dysfunction (41). Furthermore, some lncRNAs expressed in the tumor cells are affected by immune cells.…”

“…TEx from head and neck cancers induce a CD27 ‐ /CD28 ‐ suppressive phenotype in CD8 T cells . Interestingly, other suppressive CD8 T cell populations are induced by hepatocellular carcinomas and prostate tumours, including exhausted (PD1 + /TIM3 + ) and inhibitory (CTLA4 + ) cells which contribute to the suppressive tumour microenvironment by exosome/IL‐35 production . Thus, atypical suppressive CD8 T cell populations contribute to tumour immune escape by secretion of immunoregulatory exosomes.…”
