Experimental keratitis in rabbits by human HSV‐1 variants: Prevention and treatment

Abstract: The efficacy of different therapies and vaccine preparations was assessed for treating or preventing herpetic ocular keratitis induced by experimental inoculation in rabbits with two HSV-1 variants that display different pathogenetic potential. Early administration of acyclovir (ACV) promoted fast healing and prevented neurologic involvements: alpha-interferon (alpha-IFN) was less efficient than ACV; combined therapy with both drugs increased the antiviral effects. In an attempt to prevent the disease, rabbits… Show more

“…As previously reported [8], the immunization of rabbits with gBl-s associated with an immunologic adjuvant provided a high degree of protection against herpetic disease, whatever the adjuvant utilized: in fact, an aluminum hydroxide solu tion was used in this case, while previous experiments were performed using Freund's complete adjuvant. In this trial, gBl-s vaccination proved very effective, both in preventing CNS involvement and animal death as well as in reducing the severity and time course o f LV herpetic keratitis.…”

“…Fifty microlitcrs o f virus suspension (containing I05 PFU) was introduced in the con junctival sac and gently massaged onto the eye with the eyelids [7], For 16 days or until the animals died, eye scores were read at 2-day intervals, using a cobalt-filtered blue light on eyes previously stained with fluorescein. Corneal lesions were graded on a scale o f 0 to +5 according to the surface area involved, as previously described [8], The eyes were photographed with a Contax camera equipped with a macrolens.…”
