Expression of Fibronectin Isoforms Bearing the Alternatively Spliced EIIIA, EIIIB, and V Segments in Corneal Alkali Burn and Keratectomy Wound Models in the Rat

Abstract: Deposition of EIIIA+ and EIIIB+ FNs in the BMZ of the keratectomy wound occurs more slowly than deposition of V+ and total FNs. EIIIA+ FN is expressed in a distribution that overlaps with that previously described for the alpha 9 integrin subunit following corneal debridement, suggesting that EIIIA-alpha 9 interactions could occur during corneal wound healing. In contrast, the relative lack of FN deposition in alkali burn wounds suggests that proteolytic degradation of FN may occur; and this, along with impair… Show more

“…Muro et al found that mice without the EDA exon in the FN protein displayed abnormal skin wound healing, whereas mice having constitutive inclusion of the EDA exon showed a major decrease in the FN levels in all tissues [19]. In corneal alkali burn and keratectomy wound, it is suggest that Fn-EIIIA play a role in the high prevalence of recurrent epithelial erosions in alkali-wounded corneas [20]. In our study, the expressions of EIIIA in skin were gradually increased and displayed linear relationship with the injury time prolonging, as a hint for skin wound estimation.…”

“…Alkali burn was performed by touching a 4.0-mm-diameter applicator (a piece of sterile gauze) saturated with 1mol/l sodium hydroxide on the corneas of the left eyes of the two groups for 30 seconds. The ocular surface was then rinsed with 30ml sterile sodium chloride [2,[9][10][11].…”

“…Enhanced alternative splicing of Fn1 variants were observed in cutaneous wounds, [24][25][26] corneal wounds, [27][28][29][30] gastric ulcer healing, 31 liver wound repair 32 and hepatic fibrogenesis, 33 myocardial infarction, 34 acute major trauma, 35 proliferative glomerulonephritis, 36,37 and peripheral nerve injury. 38 Assays of the plasma fibronectin and fetal fibronectin variants have been applied to the clinical diagnosis and therapeutic administration of fibronectin to human diseases.…”
