Fluorescence Techniques Using Dehydroergosterol to Study Cholesterol Trafficking

Abstract: Cholesterol itself has very few structural/chemical features suitable for real-time imaging in living cells. Thus, the advent of dehydroergosterol [ergosta-5,7,9(11),22-tetraen-3β-ol, DHE] the fluorescent sterol most structurally and functionally similar to cholesterol to date, has proven to be a major asset for real-time probing/elucidating the sterol environment and intracellular sterol trafficking in living organisms. DHE is a naturally-occurring, fluorescent sterol analog that faithfully mimics many of the… Show more

“…The result for cholesterol was consistent with previously published data ( 41 ). Thus, similar to other fl uorescent sterols ( 15,16,42 ), DChol exhibited a somewhat higher critical concentration for micelle/microcrystal formation in aqueous buffer than exhibited by cholesterol. Nevertheless, DChol, like cholesterol and other fl uorescent sterols, was very poorly soluble in aqueous buffer.…”

“…Biotinylated C -toxin (BC ), a biotinylated perfringolysin O derivative, was prepared by Dr. Yoshiko Ohno-Iwashita (Tokyo Metropolitan Institute of Gerontology, Japan) as described earlier ( 19 ). DHE was synthesized by a modifi cation of an established procedure as described earlier ( 15 ). Concanavalin-A (con-A) sepharose 4B resin and Percoll were purchased from GE Healthcare Bio-Sciences Corp. (Piscataway, NJ); silica gel G TLC plates were from Analtech (Newark, DE).…”

“…In total, 18 subjects were evaluated. This data display a consistent spectral appearance similar to that of AGE auto fluorescence [13,14], with a peak located at 480-490 nm (Figure 2). This result indicates that an MSI camera setup capable of capturing the light above 450nm would be suitable for detecting UV induced AGE auto fluorescence.…”

“…12, 44, 45 ) and real-time imaging ( 3,4,46,47 ) revealed the presence of cholesterol-rich and cholesterol-poor microdomains in cell membranes. Cholesterol-rich microdomains in the plasma membranes of both peripheral cells ( 8,(48)(49)(50) and hepatocytes ( 1 ) serve as the major scaffolding regions for cholesterol transport proteins important in uptake and disposition of cholesterol.…”
