“…Germination was effected for 7 days (owing to the low room temperature of 16±17°C) until shoots were 0.5±1.5 cm long. The rest of the procedure is shown in Fig 1. Vicine and convicine assay Products from treatments of fababean¯our, fababean cotyledons and germination were analysed for vicine and convicine by a slightly modi®ed procedure of Jamalian et al 14 using a solvent system of isopropyl alcohol±ammonia±water (7:1:2) 15 for the development of thin layer chromatography (TLC) plates. Other solvent systems also employed by Brown and Roberts 15 were used to develop TLC plates as crosschecks for the location of vicine and convicine.…”

Vicine and convicine bands separated by the isopropyl alcohol±ammonia±water system were eluted, their UV absorption spectra were scanned on a Hitachi UV-vis spectrophotometer and their concentrations were calculated using their respective molar extinction coef®cients.…”

“…Vicine and/or convicine were detected in various species of family Fabaceae: Vicia sativa L. (Ritthausen 1876), and Vicia faba L. (Brown and Roberts 1972). Vicine was found also in Pisum sativum L. (Jamalian et al 1977) and in Lupinus albus L. (Pompei and Lucisano 1976), but were not reported in other paper (Pitz et al 1980).…”

“…However, previous studies where whole seedlings were analysed (Brown and Roberts 1972) showed no evidence for vicine catabolism during the seedling stage, further investigations are required to more clearly understand the role of these nitrogen-rich compounds during both the development and germination of faba bean seeds. The implication of these results for the breeding of faba bean cultivars with reduced glucopyranoside contents may not become fully realised until further results identifying more clearly this physiological role within the seed have been completed.…”
